摘要
目的 检测Notch1~4蛋白在肺腺癌组织中的表达,并分析其与临床特征的关系,探讨其临床意义。方法 收集肺腺癌病例,调查肿瘤分化程度、肿瘤大小、淋巴结转移和临床分期等参数。采用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连结法(免疫组织化学SP法)分别检测腺癌组织及其癌旁组织中Notch1~4蛋白的表达,应用Image-Pro Plus 6.0图像分析软件进行图像检测,以积累光密度值(IOD值)均值表示,并分析其与临床特征的关系。结果男性肺腺癌患者的Notch1、2、3蛋白IOD值显著高于女性患者(P值均〈0.05);肿瘤TNM分期Ⅱ、Ⅲ期患者显著高于TNM分期Ⅰ期患者(P值均〈0.05),TNM分期Ⅲ期患者显著高于TNM分期Ⅱ期患者(P值均〈0.05);有淋巴结转移的患者显著高于无淋巴结转移的患者(P值均〈0.05);有远处转移的患者显著高于无远处转移的患者(P值均〈0.05);不同年龄、肿瘤分化程度、肿瘤大小的患者间差异均无统计学意义(P值均〉0.05)。Notch4蛋白的IOD值在不同临床特征的肺腺癌患者间的差异均无统计学意义(P值均〉0.05)。结论 Notch受体各亚型中,Notch1、2、3参与肺腺癌的发生、发展和转移,可能具有促癌作用,Notch4蛋白的作用不确定。
Objective To investigate the expression of Notch 1 -4 proteins in lung adenocarcinoma tissues, to analyze their Correlation with clinical characteristics, and to discuss its role in the treatment of lung adenocarcinoma. Methods The cases of lung adenocarcinoma were collected. Tumor differentiation, tumor size, lymph node metastasis, and clinical stage were investigated. The expression of Notch 1 -4 proteins in lung adenocarcinoma tissue and the tissue adjacent to malignant tissues were detected by immunohistochemistry method. Quantitative analysis was done by Image-Pro Plus 6.0 image analysis software. Their relationship with clinical pathological factors were analyzed. Results Integrated option density (IOD) values of Notch 1, 2 and 3 proteins in male patients were significantly higher than those in females (all P〈0.05). The IOD values of Notch 1, 2 and 3 proteins in patients with the TNM staging Ⅱ and Ⅲ were significantly higher than those in patients with stage TNM staging Ⅰ (all P〈0.05). The IOD values of Notch 1, 2 and 3 proteins in patients with TNM staging ]H were also significantly higher than those in patients with TNM staging Ⅱ (all P〈0.05). The IOD values of Notch 1, 2 and 3 proteins in patients with lymph node metastasis were significantly higher than those in patients without metastasis (all P〈0.05). The IOD values of Notch 1, 2 and 3 proteins in patients with distant metastasis were significantly higher than those in patients without distant metastases (all P〈0.05). The IOD values of Notch 1, 2 and 3 proteins were not statistically different in patients with different ages, differentiation degrees and tumor sizes(all P〉0.05). There was no significant difference in IOD value of Notch 4 protein in different lung adenocarcinoma patients (all P〉0.05). Conclusion Notch 1-3 proteins may participate in the carcinogenesis, development and metastasis of lung adenocarcinoma, while the influence of Notch 4 is uncertain.
