壬基酚对仔鼠皮质部星形胶质细胞GFAP的影响

Effects of nonylphenol on GFAP in cerebral cortex astrocytes of newborn rats

摘要 将交配成功的31只孕鼠根据妊娠日期分层后随机分为4组:对照组,壬基酚(NP)低、中、高剂量(25、50、100 mg/kg)染毒组;NP暴露时间为受孕第6天到出生后21 d哺乳期结束。Real Time PCR和免疫组织化学法检测胶质原纤维酸性蛋白(GFAP)mRNA和蛋白的表达,并分析上述变化与NP暴露之间的关系。高剂量组21 d、60 d海马GFAP mRNA表达增加,差异有统计学意义(P<0.05),21d、60 d仔鼠皮质部位GFAP mRNA表达与NP暴露剂量呈正相关(r=0.670、0.588,P<0.05),免疫组化切片可见仔鼠皮质部位星形胶质细胞体肥大,细胞分支增多,染色较深,高剂量组GFAP阳性细胞数与NP暴露剂量呈正相关(r=0.624、0.713,P<0.05);GFAP光密度与NP暴露剂量呈正相关(r=0.733、0.679,P<0.05)。各暴露组21 d仔鼠海马GFAP阳性细胞数低于60 d(t=-5.525,P<0.05)。提示NP孕期暴露影响子代大鼠脑组织GFAP的表达,且剂量越高,GFAP表达越低。
