天麻药材中天麻素含量测定的样品制备方法比较

Comparision of Different Method for Sample Preparation and Extraction of Gastradin in Gastradia elata Bl.

目的:天麻药材中天麻素含量测定时样品的最佳制备方法。方法:采用高效液相色谱法,比较了稀乙醇回流提取法(2010年版药典天麻项下方法)、80%甲醇超声法(2010年版药典天麻祛风补片项下方法)、甲醇超声-静置-超声法(2010年版药典天麻头痛片项下方法)、稀乙醇浸渍-回流法(2010年版药典天麻钩藤颗粒项下方法)及稀乙醇索氏提取5种样品制备方法对天麻素含量测定结果的影响,并对超声法的提取溶剂和提取时间进行了考察。结果:50%甲醇超声提取1h优于其它方法。结论:本方法操作简便可行、重现性好、准确度高,可作为天麻药材中天麻素含量测定的供试品溶液制备方法。

Objective：To optimize the method for extraction of gastrodin in Gastrodia elata Bl..Methods：The five methods were investiga-ted.The first was refluxed in 49.5%-50.5%ethanol;the second was extracted in ultrasonic bath with 80%methanol;the third was extracted in ultrasonic bath for 30 minutes,soaked for 24 hours and extracted in ultrasonic bath for 30 minutes again with methanol;the forth was soaked and refluxed in 49.5%-50.5%ethanol orderly;the fifth was extracted in soxhlet’s extractor with 49.5%-50.5%ethanol.Extraction solvent and extraction time were investigated in the method of extraction in ultrasonic bath.Results：The highest ex-traction rate of gastrodin was obtained in ultrasonic bath with 50%methanol for 1 hour.Conclusion：The established method was sam-ple,reproducible and accurate.
