摘要
目的:探讨桉叶油对体外培养人绒毛膜细胞生长及分泌功能的影响。方法胰酶消化法收集正常妊娠6∽8周人绒毛膜细胞,分别用0.75、0.15、0.015、0.0015g·L-1的桉叶油作用12h、24h、48h,倒置显微镜观察细胞形态变化,活细胞计数测定细胞的生长曲线,MTT法检测桉叶油对细胞的抑制率,化学发光法检测细胞培养上清液中的雌激素、孕激素和绒毛膜促性腺激素的浓度。结果与对照组比较,质量浓度≥0.75g·L-1的桉叶油对绒毛膜细胞生长有明显的抑制作用(P〈0.05);质量浓度≤0.15g/L的桉叶油对绒毛膜细胞生长的抑制率与对照组比较无明显差异(P>0.05)。桉叶油作用绒毛膜细胞24h 时,0.75g/L桉叶油组细胞上清液中雌二醇、孕激素浓度均较对照组显著降低(P〈0.05),其余各组细胞上清液中雌二醇、孕激素浓度均与对照组比较无差异。桉叶油作用绒毛膜细胞24h时,桉叶油各组细胞上清液中绒毛膜促性腺激素浓度较对照组显著增高(P〈0.05)。结论质量浓度≥0.75g/L桉叶油对人绒毛膜细胞生长有抑制作用,质量浓度≤0.15g/L桉叶油对人绒毛膜细胞生长无抑制作用。桉叶油对体外培养的绒毛膜细胞分泌雌二醇、孕激素无明显影响,但可刺激绒毛膜细胞分泌HCG。
Objective To investigate the effect of eucalyptus on the growth and secretion of human chorionic cells. Methods Human chorionic cells were isolated from early villus of pregnancy women (6∽8 weeks) by trypsin digestionand and incubated in DMEM medium. Eucalyptus concentrations of 0.75,0.15,0.015,0.0015g/L were exposed to human chorionic cells at different time respectively and then observed cell morphologic changes by inverted microscope , the growth curve of human chorionic cells was observed by cell count. The inhibitory effect of human chorionic cells was detected by MTT assay. The cell culture supernatant of estrogen, progesterone and human chorionic gonadotropin concentrations were detected by chemilumines-cence. Results Compared with control group, the mass concentration of eucalyptus over 0.75g/L had significant inhibiting ef-fect(P〈0.05),while the concentration of eucalyptus under 0.15g/L couldn't obviously prohibit the growth of human chorionic cells compared with control group. Human chorionic cells were treated by eucalyptus for 24h , supernatant estradiol and progesterone level of the concentration of eucalyptus over 0.75g/L cell reduced observably than control group (P〈0.05). The other eucalyptus groups all showed no significant difference compared with control group. HCG concentration of the eucalyptus group was higher than control group (P〈0.05). Conclusion The concentration of eucalyptus over 0.75g/L could prohibit the growth of human chorionic cells. It shows a very substantial difference in antiproliferative and cytotoxic effect. While under the concentration of 0.15g/L eucalyptus couldn't show obviously antiproliferative and cytotoxic effect on human chorionic cells. Eucalyptus has no significant effect on cultured chorionic cells secretion of estradiol and progesterone but can stimulate the cells to secrete human chorionic HCG.
