AGT基因表观调控在原发性高血压不同中医证型中的作用研究 被引量:4

摘要 目的从表观遗传学水平阐述AGT基因与原发性高血压发生及中医各证型的相关性。方法提取各中医证型原发性高血压患者血液的总RNA,采用实时荧光定量PCR半定量法研究各对象AGT基因m RNA的表达量,同时提取所采血液的DNA,采用焦磷酸测序技术检测AGT基因启动子区部分序列的甲基化状态,比较各组甲基化情况。结果 AGT基因启动子区5个被检Cp G位点中,有2个Cp G位点甲基化指数存在差异,在正常对照组中呈高甲基化状态,而肝火亢盛证呈低甲基化状态。结论原发性高血压患者AGT基因m RNA表达量高于正常人群,且呈肝火亢盛证>阴虚阳亢证>痰湿壅盛证>阴阳两虚证排列。AGT基因甲基化修饰与原发性高血压发生有关,并与原发性高血压中医证型相关联。 Objective To clarify the association between the AGT gene with primary hypertension occuring and tradi- tional chinese medicine (TCM) syndromes of primary hypertension. Methods Extracted total RNA in blood and tested ACT gene mRNA expression of participants by real-time fluorescent quantitative PCR. Then compared the results of different groups. Extracted DNA in blood and analyzed the AGT gene methylation status in promoter region of participants by pyrosequeneing. Then compared the results of different groups. Results Two CpG sites in five CpG sites were different in the situation about AGT gene methylation status in promoter region of five groups. Control group was high methylation status ,but Sthenie liver Yang type group was low methylation status. Conclusion AGT gene mRNA expression of primary hypertension patients were higher than normal people. AGT gene mRNA expression in different syndromes from high to low is Sthenic liver Yang type, Sthenic Yang and Yin defieiancy type, Stagnation of phlegmatic hygrosis type, both Yang and Yin deficiency type. There is association between AGT gene methylated modification with primary hypertension and TCM syndromes of primary hypertension.
