猪骨渣中可溶性钙的提取及蛋白肽螯合钙的制备 被引量:4

摘要 [目的]首先优化出猪骨渣中可溶性钙提取的工艺,然后在此基础上将其制备成蛋白肽螯合钙。[方法]首先研究影响提取猪骨渣中可溶性钙的几个因素,包括酸种类、酸用量、液料比、提取时间和提取温度,并通过正交试验设计与分析优化最佳工艺参数;然后研究影响蛋白肽螯合钙制备的几个因素,包括配比、p H值、反应温度和反应时间。[结果]猪骨渣中可溶性钙提取的最佳工艺参数为酸种类盐酸、盐酸用量(V盐酸∶m猪骨渣,下同)2m L/g、液料比(V提取液∶m猪骨渣,下同)10m L/g和提取时间40min,在此条件下实际得率为15.01%;蛋白肽螯合钙制备的最佳工艺参数为配比(V可溶性钙提取液∶V蛋白肽酶解液,下同)1∶4,p H值8,反应温度50℃和反应时间10min,在此条件下实际螯合率为95%。[结论]采用盐酸提取猪骨渣中可溶性钙及在此基础上将其制备成蛋白肽螯合钙都是可行的。 [ Objective] The process of extracting soluble calcium from pig bone residue was optimized. And then chelated peptide calcium compound was prepared using extracting soluble calcium. [ Method] Factors affected the extraction process including acid type, amount of hydrochloric acid, solvent to material ratio, extraction temperature and extraction time, was selected and processing parameters of were optimized by the orthogonal experimental design. Finally, the affected factors in the preparation of peptide chelated soluble calcium compound including ratio, pH value, reaction temperature and reaction time, were studied. [ Results ] The best extraction process parameters of soluble calcium from pig bone residue were hydrochloric acid 2mL/g, solvent to material ratio 10mL/g and extraction time 40min. Under the optimized extraction conditions, the extraction yield was 15.01%. The best process parameters were: ratio of soluble calcium and peptide enzyme solution were 1 : 4, pH value 8, reaction temperature 50℃ and reaction time 10min. Under the optimized chelation conditions, the actual chelating rate was 95%. [ Conclusion] The extraction of soluble calcium from pig bone residue with hydrochloric acid and the preparation of peptide chelated soluble calcium compound using soluble calcium from pig bone residue were feasible.
