摘要 目的:建立固相萃取一高效液相色谱法测定婴幼儿食品中的VA、VD、VE含量的方法。方法:在已有研究基础上对样品前处理方法进行了优化和改进,皂化后利用正己烷:甲基叔丁基醚(v:v=1:1)进行提取,PLS小柱萃取分离,50%丙酮正己烷洗脱,甲醇定容后高效液相色谱仪梯度洗脱检测。结果VA、VD、VE在0.1~10μg/mL浓度范围内相关系数均为0.999以上,回收率均在90%以上,相对标准偏差小于4%。结论:方法快速、灵敏、准确,前处理简单易行,可用于婴幼儿食品中VA、VD、VE的同时测定。 Object: A solid phase extraction followed by high performance liquid chromatographic method was developed for the determination of vitamin A, D and E in infant food. Method: The research optimized and improved the sample pretreatment on the basis of existing study. N - hexane and methyl tert - butyl ether( v : v = 1 : 1 ) were used to extract active ingredients after saponification, PLS solid phase extraction column was used and 50% acetone and n - hexane was eluted solution. The residue was fihrated with membrane and reserved for analysis after dissolved in methanol prior to analysis on a high performance liquid chromatography equipped with a UV detector by gradient elution. Results : The correlation coefficients of the method were all above 0. 999. The average of recoveries of target drugs were better than 85% with relative standard deviations(RSDs) less than 4%. Conclusion: Because of the advantages of good linearity, low limit of detection, high precision and rapidity, this method is suitable for the determination of vitamin A, D and E in infant food.
