摘要
目的:研究咖啡碱与落新妇苷对小鼠体重、脂肪沉积和脂类代谢酶的影响。方法:将70只雌性昆明小鼠随机分成7组,即对照组(I)、0.06%咖啡碱(II)、0.4%落新妇苷(III)、0.03%咖啡碱+0.2%落新妇苷(IV)、0.03%咖啡碱+0.4%落新妇苷(V)、0.06%咖啡碱+0.2%落新妇苷(VI)、0.06%咖啡碱+0.4%落新妇苷(VII),喂养12周。投喂结束后,摘取小鼠各脏器以及腹腔内脂肪并称重,测定血中和肝脏中脂类含量以及肝脏中肉毒碱脂酰转移酶(CAT)、脂肪酸合成酶(FAS)的活性。结果:与对照组相比,在喂养第12周,V、VI、VII组小鼠体重增加量显著减少(P<0.05)。V、VI组的腹腔内脂肪质量显著降低(P<0.05),VII组极显著降低(P<0.01)。V、VII组小鼠的血中总胆固醇(TC)和VII组的甘油三酯(TG)浓度显著降低(P<0.05)。VII组肝脏TC以及V、VI、VII组的TG含量明显减少(P<0.05)。VII组肝脏中FAS活性显著降低(P<0.05)。结论:咖啡碱与落新妇苷组合能通过调节FAS活性,减少脂肪合成,降低小鼠血清和肝脏中的脂类含量,抑制脂肪沉积,减少体重增加,且比单独投喂效果明显,表明咖啡碱与落新妇苷对降脂减肥有一定的协同作用。
Objective: To investigate the effects of caffeine and astilbin on body weight, fat accumulation and fat metabolic enzymes in mice. Methods: Seventy female Kunming mice were randomly divided into 7 groups, fed diets containing none(Ⅰ), 0.06% caffeine(Ⅱ), 0.4% astilbin(Ⅲ), 0.03% caffeine + 0.2% astilbin(Ⅳ)., 0.03% caffeine + 0.4%astilbin(Ⅴ), 0.06% caffeine + 0.2% astilbin(Ⅵ), 0.06% caffeine + 0.4% astilbin(Ⅶ) for 12 weeks respectively. Organs and intraperitineal adipose tissues(IPAT) were weighted at the end of experiment. Serum lipids concentrations and liver lipids levels were measured. The activities of fatty acid synthese(FAS) and carnitine acyltransferase in liver were determined. Results: At the 12 th week, the body weight gain was decreased significantly(P〈0.05) by Ⅴ, Ⅵ and Ⅶ groups. The IPAT weights of Ⅴ and Ⅵ groups reduced significantly(P〈0.05), and Ⅶ group reduced very significantly(P〈0.01). Serum TC concentration was significantly reduced(P〈0.05) by Ⅴ and Ⅶ groups, and TG concentration was significantly reduced(P〈0.05) by Ⅶ group. Ⅶ group significantly decreased TC, and Ⅴ, Ⅵ and Ⅶ groups reduced TG in liver. Ⅶ group significantly reduced FAS activity(P〈0.05). Conclusions: Caffeine and astilbin combination can reduce FAS activity, inhibit fat synthesis, and reduce the lipids level in serum and liver. It causes the suppressive effect on fat accumulation and body weight gain, and than their single effect more apparent. Moreover, it was indicated that caffeine and astilbin was synergistic in anti-obesity effect.
