塑料中PAEs环境污染物向食品模拟物中迁移及对CYP3A4和芳香化酶的影响 被引量:5

Migration of Phthalate Esters(PAEs) in Food Simulacrum and Their Effect on the Expression of Enzymes(CYP3A4 and Aromatase)

摘要 目的:探讨塑料中PAEs向食品模拟物中迁移的规律,阐明PAEs对代谢酶CYP3A4和芳香化酶表达水平的影响。方法:利用GC-FID建立不同食品模拟物中5种PAEs类化合物检测方法,检测不同食品模拟物中PAEs类化合物的迁移量。采用q RT-PCR技术检测PAEs类化合物对肝脏代谢酶CYP3A4和睾丸间质细胞中芳香化酶m RNA转录水平的影响,采用荧光免疫组织化学技术分析对其蛋白表达的影响。结果:不同食品模拟物中5种PAEs类化合物迁移量不同,其中以异辛烷中迁移量最大,而且迁移量随着时间的延长而增加。PAEs类化合物显著诱导肝脏中代谢酶CYP3A4基因表达,而对睾丸中芳香化酶基因表达具有极显著的抑制作用。结论:PAEs类化合物在不同极性食品中迁移量不同,脂溶性越高迁移量越大;PAEs类化合物对肝脏CYP3A4和睾丸芳香化酶表达有显著影响,影响体内类固醇激素的浓度,从而表现出生殖内分泌毒性。 Objective: The study was designed to investigate the rule of transferring phthalate esters(PAEs) into four food simulates. Then the aim of the study was to clarify the effect of PAEs chemicals on expression levels of CYP3A4 and aromatase. Methods: The GC-FID method was used to determine the five PAEs chemicals in these food simulates.And the transfer amounts of the five PAEs chemicals were detected. Then the expression levels of CYP3A4 and aromatase m RNA were also detected by q RT-PCR technology and the protein levels of them were analyzed according to the results of Immune fluorescence staining. Results: The results showed that the different amounts of the five PAEs chemicals were transferred into the different food simulates. And the most amount of the five PAEs chemicals was detected in the food simulate of isooctane. Then the transferred amounts of the five PAEs increased as the time prolonged. On the other hand, the expression of CYP3A4 was significantly increased in liver, while the expression of aromatase was significantly decreased in testis. Conclusion: The higher lipid solubility of food simulates, the more amount of PAEs chemicals transferred. And PAEs chemicals significantly affected the expression of liver CYP3A4 and testis aromatase, which would affect the metabolism of steroid hormone and show the reproductive toxicity.
