鞘内注射TRESK基因重组腺病毒对坐骨神经分支选择性损伤大鼠的镇痛作用 被引量:1

Effects of recombinant adenovirus vector containing TRESK gene by intrathecal injection on hyperalgesia of sciatic nerve branch selective injury rats

摘要 目的研究鞘内注射TRESK基因重组腺病毒(p Ad/CMV/V5-DEST-TRESK)对坐骨神经分支选择性损伤(SNI)大鼠痛觉过敏的影响。方法雄性SD大鼠36只,随机分为6组(n=6):Ⅰ组为空白对照组;Ⅱ组为假手术组;Ⅲ组为SNI模型组;Ⅳ组大鼠制备SNI模型后鞘内单次注射生理盐水25μl;Ⅴ组和Ⅵ组大鼠制备SNI模型后,鞘内分别单次注射滴度为1.31×109TU/ml的阴性腺病毒和p Ad/CMV/V5-DEST-TRESK。各组大鼠于术前1 d及术后1 d、3 d、5 d、7 d、14d分别测定机械痛阈和热痛阈;术后14 d检测L4,5术侧背根神经节(DRG)的TRESK蛋白表达。结果Ⅲ组、Ⅳ组、Ⅴ组、Ⅵ组术后1 d、3 d、5 d、7 d、14 d的机械痛阈明显低于Ⅰ组(P〈0.05),Ⅲ组、Ⅳ组、Ⅴ组5 d、7 d、14 d的机械痛阈明显低于Ⅵ组(P〈0.05);与术前比较,Ⅲ组、Ⅳ组、Ⅴ组、Ⅵ组术后1-14 d的机械痛阈明显降低(P〈0.05)。Ⅲ组、Ⅳ组、Ⅴ组、Ⅵ组L4,5术侧DRG的TRESK蛋白表达明显低于Ⅰ组(P〈0.05),Ⅵ组L4,5术侧DRG的TRESK蛋白表达明显高于Ⅲ组、Ⅳ组、Ⅴ组(P〈0.05)。结论鞘内注射p Ad/CMV/V5-DEST-TRESK可上调SNI大鼠DRG的TRESK蛋白表达,减轻SNI大鼠的痛觉过敏。 Objective To investigate the effects of recombinant adenovirus vector containing TRESK gene( pAD/CMV/V5 -DEST-TRESK)by intrathecal injection on hyperalgesia of sciatic nerve branch selective injury( SNI)rats. Methods Thirty six SD male rats were randomly divided into six groups,six rats in each groups:control group( Group I);sham operation group( GroupⅡ);SNI group( GroupⅢ). SNI rats were given respectively single intrathecal injected 25μl physiological saline. The negative adenovirus and pAD/CMV/V5-DEST-TRESK in Group Ⅳ,Group Ⅴ and GroupⅥ,the titer of adenovirus was 1. 31 × 109 TU/ml. Rats in each groups were determined by mechanical threshold (MWT)and heat pain threshold respectively(TWL)at 1d before operation and 1,3,5,7,14 d after operation. The protein of TRESK in the side of L4,5 DRG were determined at 14 d after operation. Results The MWT were significantly lower in GroupⅢ,Ⅳ,ⅤandⅥthan in Group I at 1d before operation and 1,3,5,7,14 d after operation( P ﹤0. 05). The MWT were significantly lower in GroupⅢ,ⅣandⅤthan in GroupⅥat 5,7,14 d after operation( P ﹤0. 05). The MWT in GroupⅢ,Ⅳ,ⅤandⅥwere significantly lower at 1,3,5,7,14 day after operation than at 1 day before operation( P ﹤0. 05). The protein of TRESK in DRG in GroupⅢ,Ⅳ,ⅤandⅥwere significantly lower than in Group I ( P ﹤0. 05). The protein of TRESK in DRG in GroupⅥwere significantly higher than in GroupⅢ,ⅣandⅤ( P ﹤0. 05). Conclusion Intrathecal injection with the recombinant adenovirus vector containing TRESK gene( pAD/CMV/V5-DEST-TRESK )can up-regulate the protein of TRESK in DRG and alleviate the hyperalgesia of SNI rats.
