黄芪注射液对大鼠急性脊髓损伤的神经保护作用 被引量:8

摘要 目的探讨黄芪注射液对急性脊髓损伤大鼠的神经保护作用及其机制。方法将40只成年健康SD大鼠随机分为4组,每组10只。采用改良ALLEN氏重物打击法建立脊髓损伤大鼠模型。A组给予黄芪注射液2 m L/(kg·d),连续给药14 d。B组给予黄芪注射液0.5 m L/(kg·d),连续给药14 d。C组术后立即腹腔注射甲基强的松30 mg/kg,24 h内每隔4 h后重复给药1次。D组给予灭菌生理用水0.5 m L/(kg·d)连续14 d。各组用药后1、5、10、14 d分别进行Rivlin斜板实验对大鼠脊髓功能的行为学评分进行评价,采用改良Gale联合行为评分(CBS)测定大鼠脊髓损伤后的神经学方面功能,在给药后48 h测定脊髓损伤组织的重量,并根据公式计算脊髓损伤组织的含水量。结果 A组各时间点Rivlin斜板度数明显高于B组和D组,差异有统计学意义(P<0.05);A组效果明显优于D组和B组。与D组相比,A组大鼠损伤脊髓的神经功能得到改善,CBS明显提高。根据公式计算脊髓损伤组织的含水量,D组和B组的含水量明显高于A组,而A组和C组的差异无统计学意义。结论高剂量黄芪注射液能够改善大鼠损伤脊髓的神经功能,降低脊髓损伤组织的含水量,从而减轻组织的水肿。 Objective To explore the neuroprotective function of astragalus injection on treat- ment of rats with acute spinal cord injury. Methods 40 health male SD rats were randomly divided into 4 groups, 10 rats in each group. The model of rats with acute spinal cord injury was established according to Allen's methods. The group A was injected with a dosage of 2 mL/(kg ·d). The group B was injected with a dosage of 0.5 mL/( kg · d). The group C was injected with 30 mg/kg methyl- prednisolone and group D was injected with a dosage of 0.5 mL/(kg· d) saline. Inclined plane test was performed to evaluate the effects of exercise function 1, 5, 10 and 14 days after model creation. The neurological function of injured tissue of the spinal cord was evaluated by the Combined Behavioral Score (CBS). The samples taken from the spinal cord injury sites were detected in the water content of spinal tit^sue after injection of 48 hours. Results The scores of inclined plane test in group A were significa:ady higher than group B and group D ( P 〈 0.01 ). The neurological function of the injured spinal cord in the group A was significantly better than the group D (P 〈 0.05). The water content of the injured tissue of the spinal cord in group D and group B were higher than group A, but there was no significant difference between group C and group A. Condusion High dose of astragalus injection can improve the neurological function of the injured spinal cord, decrease the water content of spinal tissue and reduce edema.
