亚甲基四氢叶酸还原酶基因多态性与甲氨蝶呤不良反应的相关性 被引量:7

摘要 目的探讨亚甲基四氢叶酸还原酶(MTHFR)基因第677位点多态性与大剂量甲氨蝶呤(HD-MTX)化疗不良反应的相关性。方法用聚合酶链反应-芯片杂交技术检测MTHFR C677T多态性,酶放大免疫分析法测定患者血药浓度,记录HD-MTX化疗后相关不良反应。结果 MTHFR677 CT+TT基因型者发生口腔黏膜损害的风险较CC型者显著性增高(P<0.05)。结论 MTHFR C677 T基因多态性与HD-MTX化疗所致口腔黏膜损害存在相关性。 Objective To study the association of methylenetetra-hydrofolate reductase ( MTHFR ) gene C677 T polymorphism and ad-verse drug reaction ( ADR ) by high -dose methotrexate ( HD -MTX) .Methods Polymerase chain reaction-microarray hybridiza-tion technology was used to detect the polymorphism of MTHFR C677 T.Enzyme multiplied immunoassay was used to monitor dynamic serum concentration of patients.Methylenetetrahydrofolate related ADR were recorded.Results The probability of mucositis was higher in carriers of MTHFR 677 CT+TT genotype as compared with patients with 677 CC genotype ( P 〈0.05 ).Conclusion The MTHFR C677 T polymorphism may contribute to the mucositis induced by HD-MTX chemotherapy.
