摘要
目的观察尤瑞克林对脑缺血再灌注大鼠血管内皮生长因子(VEGF)和碱性成纤维细胞生长因子(b FGF)表达的影响。方法 SD大鼠随机分为假手术组、模型组和实验组。线栓法制作大脑中动脉脑缺血再灌注模型,实验组于再灌注后5 min静脉注射尤瑞克林3.5×10-3PNAU·kg-1。缺血2 h后再灌注48 h,断头取脑,免疫组织化学法和PCR法检测VEGF和b FGF的表达。结果与模型组相比,实验组大鼠脑组织中VEGF和b FGF的表达水平明显增高,差异均有统计学意义(P<0.05)。结论尤瑞克林对大鼠脑缺血再灌注损伤的保护作用可能与促进VEGF和b FGF的表达有关。
Objective To investigate the effects of urinary kallidinoge-nase on the expression of vascular endothelial growth factor ( VEGF ) and basic fibroblast growth factor ( bFGF ) in rats with focal cerebral ischemia-reperfusion.Methods Rats were randomly assigned into three groups: sham operation group ( sham group ) , focal cerebral ischemia-reperfusion group ( model group ) and urinary kallidinogenase group ( test group ) .The middle cerebral artery occlusion reperfusion model was made by the suture method (ischemia for 2 hours, and reper-fusion for 48 hours).The rats in the test group were injected with urinary kallidinogenase 3.5 ×10 -3 PNAU.kg -1 after making a model.The level of VEGF and bFGF expression were measured by immunohistochemistry and PCR.Results The expression of VEGF and bFGF in the rats of test group were higher than those in model group ( P〈0.05 ).Conclusion Urinary kallidinogenase can relieve the brain damage partly by enhancingVEGF and bFGF synthesis in brain.
