摘要
目的寻找食管鳞癌早期诊断、疗效评估、预后监测的生物学标记物。方法应用双向凝胶电泳分析10例患者食管鳞癌组织和相应癌旁组织的蛋白表达谱,并对差异表达蛋白质进行质谱鉴定。结果共鉴定出29个差异表达蛋白,包括核不均一核糖核蛋白F等12个上调蛋白和人类蛋白酶体激活剂PA28β等17个下调蛋白。结论食管鳞癌组织与相应癌旁组织的蛋白表达谱的表达具有显著差异性,表达差异的蛋白有可能成为食管鳞癌潜在的肿瘤标志物。
Objective To identify biomarkers for early detection,prognostic evaluation,and recurrence monitoring of esophageal squamous cell carcinoma(ESCC).Methods Differential expression profiles of 10 ESCC core tissues and paired normal esophageal epithelial tissues were detected by two-dimensional gel electrophoresis(2-DE),matrix-assisted laser desorption/ionization-time-of-flight/time-of-flight mass spectrometry(MALDI-TOF/TOF-MS).Results There were 12 proteins,including Heterogeneous nuclear ribonucleoprotein F(hnRNP F),were upregulated in ESCC tissues;17proteins,including proteasome activator 28-beta subunit(PA28β),were downregulated.Conclusion Compared with paired normal esophageal epithelial tissues,the protein profile of ESCC core tissues showed part of difference,and the differentially expressed protein could be the potential tumor biomarker of ESCC.
