摘要
目的 建立RP-HPLC法测定王氏保赤丸中盐酸小檗碱的含量.方法 采用反相高效液相色谱法测定,以Diamonsil-C18柱(250 mm×4.6 mm,5μm)为色谱柱,以乙腈-0.05 mol/L磷酸二氢钾(28∶72)磷酸调pH3.0为流动相.柱温:室温,流速:1.0 mL/min,检测波长:349 nm.结果 盐酸小檗碱在0.20~1.20μg范围内的线性关系良好,其线性方程为A=3673.7m+5.5251,r=0.9999,平均回收率为100.49%,RSD=1.46%(n=9).结论 RP-HPLC法测定王氏保赤丸中盐酸小檗碱的含量线性关系良好、精密度较好、准确度较高、稳定性良好,可用于王氏保赤丸的质量控制.
Objective To establish a method for the determination of the berberine hydrochloride in Wangshibaochi Pill by RP-HPLC. Methods Diamonsil-C18 column(250 mm×4.6 mm, 5 μm), acetonitrile-0.05 mol/LKHz PO3 (28 : 72) as mobile phase, adding phosphoric aci to make the pH value 3. 0. The column temperature was room temperature, the flow rate was 1. 0 mL. rain 1, and the detection wavelength was 349 nm. Results The concentration of the berberine hydrochloride was linearly related to its peak area in the rang of 0.20 ~1.20μg, the linear regression equation is A =3 673.7m+5.525 1, r = 0.999 9. The average recovery was 100.49%,withRSD of 1.46% (n = 9). Conclusion This method is linear,accurate,precise and stabile, it can be used for quality control of Wangshibaochi pills.
