摘要
目的:建立维药睡莲花药材中没食子酸的含量测定方法,为其质量控制提供科学依据。方法:采用HPLC法测定,色谱条件:Agilent Eclipse XDB-C18色谱柱(250mm×4.6mm,5μm),流动相为甲醇:0.05ml/l磷酸二元梯度洗脱,流速:1.0ml/min,柱温:30℃,检测波长为273nm。结果:没食子酸在0.00196~0.0784mg/ml的范围内与峰面积呈良好的线性关系(r=0.9999),加样回收率为98.12%,RSD为1.79%(n=6);样品溶液在32h内稳定。结论:该方法操作简便、重复性好,可用于规范和控制维药睡莲花药材的质量。
Objective To establish the determination method of Nymphaea candida Presl.for determining gallic acid with HPLC. Method The HPLC separation was performed on a Agilent Eclipse XDB -C18 column (250mm×4. 6mm,5μm)in gradient elute mode with a mixture consisting of methol and 0. 5ml/L phosphate acid solution at 30℃,the flow rate was 1. 0ml/min,the detection wavelength was set at 273nm.Result The calibration curve of gallic acid was linear in the range of 0. 00196 -0. 0784mg/ml (r=0. 9999);the average recovery of quercitrin was 98. 12%(RSD=1. 79%,n=6),and the properties of sample solution kept steady within 32 hours.Conclusion The method is simple,accurate and reproducible,and can be used for the quality control of Nymphaea candida Presl.
