C-Fos与HR-HPV病毒负荷量在宫颈病变及宫颈癌中表达的相关性研究 被引量:5

THE RELATIONSHIP BETWEEN THE C-FOS AND VIRAL LOAD OF HR-HPV IN THE CERVICAL LESIONS AND CERVICAL CANCER

摘要 目的探讨原癌基因蛋白(C-Fos)在宫颈病变组织及宫颈癌中的表达及与高危人乳头瘤病毒(HR—HPV)感染的相关性。方法收集2012--2014年就诊于唐山市妇幼保健院125例慢性宫颈炎、宫颈上皮内瘤变及宫颈癌患者的宫颈组织标本及相应宫颈脱落细胞标本,采用免疫组化(SP)法检测宫颈组织C-Fos基因蛋白的表达,同时采用杂交捕获法(Hc-Ⅱ)检测相应宫颈脱落细胞中HR—HPV病毒负荷量,分析二者在宫颈病变及宫颈癌组织中表达的相关性。结果125例标本中HRHPV阳性81例,其中1≤病毒负荷量(RLU/CO)≤100共40例,100~1000共33例,〉1000为8例;C-Fos在慢性宫颈炎、CINI、CINⅡ、CINⅢ及宫颈癌中阳性表达率分别为0%,33.3%,42.9%,52.2%,67.5%,经Spearman秩相关分析显示,HR-HPV病毒负荷量与C-Fos表达有相关性(r=0.533,P=0.000)。结论随宫颈病变程度进展,HR-HPV病毒负荷量与C-Fos表达率均逐渐升高,且二者具有相关性.有望将二者联合检测应用于临床。进而预测宫颈病变及宫颈癌的痛变程度。 Objective To study the relationship of the viral load of HR- HPV and C- Fos during the de- velopment of cervical lesions and cervical carcinoma. Methods A total of 125 tissues specimens which had been detected the viral load of HPV by HC- II, including chronic cervicitis, cervical intraepithelial neopla- sia (CIN) and Cervical cancer underwent immunuhisto - chemistry to detect the expression of C- Fos, and evaluated their correlations. Results Totally 81 out of 125 samples were HR- HPV DNA positive, the viral load of HPV were 1≤RLU/CO≤100(40/125),100 -1000 (33/125),≥1 000 (8/125) , the positive expression rate of C - Fos in chronic cervieitis, CIN I -Ⅲ and cervix, cervical were 0%, 33. 3%, 42.9%, 52.2 %, 67.5 % respectively. Statistical analysis for the association of C - Fos and the viral load of HPV revealed that the expression levels of C- Fos and the viral load of HR- HPV showed a significant correlation (r=0. 533,P = 0. 000). Conclusions UP - regulation of C - Fos and the viral load of HR - HPV show a significant correlation during the pathogenesis of CIN and cervical squamous, to detect both of them is helpful to calculate the tendency of cervical lesions and cervical carcinoma changes.
