桑青枯雷尔氏菌gsp C和gsp M基因的获取与序列分析

摘要 Ⅱ型分泌系统广泛存在于革兰氏阴性细菌中,转膜蛋白基因簇是其分泌途径的中心。利用PCR扩增和测序,获得了桑青枯雷尔氏菌MR111的gsp C基因707 bp和gsp M基因575 bp的序列,SMART软件预测gsp C基因编码的7~29位氨基酸为跨膜结构域,羧基末端无PDZ结构,gsp M的胞质结构域有2个α螺旋和4个β折叠。使用gsp C和gsp M的保守序列组成的联合矩阵构建系统发育树,结果表明,桑青枯雷尔氏菌MR111与罗尔斯顿氏菌和雷尔氏菌属优先聚合在同一分支,伯克氏菌和伯克霍尔氏菌位于较远分支。 The type Ⅱ secretion system widely exists in gram negative bacteria, and the probable general secretory pathway transmembrane protein clusters is the center of it. Using PCR amplification and sequencing, the sequence of gsp C and gsp M genes of mulberry Ralstonia solanacearum MR111, 707, 575 bp in length, respectively, were obtained. The structural domain analysis by SMART showed that there was one transmembrane domain(7- 29 aa)but no C- terminal PDZ inside the gsp C gene, and there were 2 alpha helix and 4 β folding in the cytoplasmic domain of gsp M gene. The results of phylogenetic tree based on the combined matrix of conserved sequences of gsp C and gsp M showed that Ralstonia solanacearum MR111, Ralstonia pickettii and other Ralstonia sp. preferred polymerization at the same branch, Cupriavidus sp. and Burkholderia sp. were in the distant branch.
