摘要
替诺福韦与氯甲基碳酸异丙酯经酯化、成盐可制得富马酸替诺福韦酯,但重结晶过程中易转变为1:0.5(碱:酸)的盐,通过碱化、再成盐步骤制得了高纯度富马酸替诺福韦酯(1:1),总收率51.8%(以替诺福韦计),纯度99.7%。
Tenofovir disoproxil fumarate was synthesized from tenofovir via esterification and salt formation. During the process of recrystallization, the salt was found to turn into 1 : 0.5 (base : acid) of salt. The high purity product (1 : 1) was obtained through alkalization and salt formation again with an overall yield of 51.8 % (based on tenofovir) and an HPLC purity of 99.7 %.
