摘要
目的探讨通解口服液对多器官功能障碍综合征(MODS)大鼠血清和回肠组织中IL-6、TNF-α的影响。方法将48只雄性SD大鼠随机分为空白组、模型组、阳性组、低剂量组、中剂量组和高剂量组,共6组,每组8只。各组均预先灌胃给予相应的干预措施,2天后,除空白组外,其余各组舌下静脉注射8μg/g脂多糖(LPS)复制MODS大鼠模型。观察大鼠生命体征(心电图、体温、呼吸幅度和频率)、血清指标(AST、ALT、CK、Cr)及血清和回肠组织IL-6、TNF-α含量。结果与空白组比较,模型组大鼠心率、体温、呼吸频率、血清指标(AST、ALT、CK、Cr)及血清和回肠组织TNF-α、IL-6含量差异有统计学意义(P<0.05,P<0.01)。与模型组比较,阳性组大鼠心率、体温差异有统计学意义(P<0.05);低、中、高剂量组大鼠心率差异有统计学意义(P<0.05,P<0.01)。与模型组比较,阳性组呼吸频率、血清指标及回肠组织中IL-6含量差异有统计学意义(P<0.05,P<0.01);高剂量组呼吸频率、血清指标、血清及回肠组织中TNF-α、IL-6含量差异有统计学意义(P<0.05,P<0.01);中剂量组血清指标、血清及回肠组织IL-6含量差异有统计学意义(P<0.05,P<0.01);低剂量组Cr水平差异有统计学意义(P<0.01)。与低剂量组比较,高剂量组血清ALT、AST、TNF-α、IL-6及回肠组织中IL-6含量,中剂量组的血清ALT含量差异有统计学意义(P<0.05,P<0.01)。与阳性组比较,低剂量组AST、ALT含量,高剂量组血清及回肠组织中IL-6含量差异有统计学意义(P<0.05,P<0.01)。结论通解口服液可有效降低脂多糖诱导的MODS大鼠血清及回肠组织中IL-6、TNF-α水平,且与其剂量具有相关性。
Objective To investigate the effect "Tongjie Oral Solution" on IL-6 and TNF-α in serum and the ileum of rats with multiple organ dysfunction syndrome (MODS). Methods Fourty-eight male SD rats were randomly divided into 6 groups: control group, model group, positive control group, low dose treatment group, medium dose treatment group and high dose treatment group, with 8 rats in each group. Intragastric administration in advance, 2 days later, intravenous inject Lipopolysaccharide (LPS) of 8mg/kg to build rat models. The vital signs such as electrocardiogram, temperature, breathing extent and respiratory rate; serum indexes such as AST, ALT, CK and Cr; levels of IL-6 and TNF-αin the serum and ileum were detected. Results There were significant differences in the heart rate, temperature, respiratory rate, serum indexes (contents of AST, ALT, CK and Cr in the serum), and contents of TNF-α and IL-6 between the blank group and model group ( P 〈 0.05, P 〈 0.01 ). There were the significant differences in the heart rate and temperature between the model group and positive control group ( P 〈 0.05 ) ; There were the significant difference in the heart rate among the low-, medium- and high-dose treatment groups ( P 〈 0.05, P 〈 0.01 ). There were significant differences in the respiratory rate, serum indexes and IL-6 level in the ileum between the model group and positive control group (P 〈0.05, P 〈0. 01 ) ; there were significant differences in the respiratory rate, serum indexes, TNF-α level and IL-6 level in the serum and ileum between the model group and high- dose treatment group ( P 〈 0.05, P 〈 0.01 ) ; there were significant differences in the serum indexes, IL-6 in the serum and ileum between the model group and medium- dose treatment group ( P 〈 0.05, P 〈 0.01 ) ; There was a significant difference in the Cr level between the model group and low-dose treatment group (P 〈 0.01 ). There was a significant difference in the levels of ALT, AST, TNF-α and IL-6 in the serum and IL-6 in the ileum between the low and high-dose treatment groups, and there was a significant difference in the ALT level between the low and medium-dose treatment groups (P 〈 0.05, P 〈 0.01 ). There were significant differences in the contents of AST and ALT bwteen the positive control group and low-dose treatment group, and with significant differences in the IL- 6 of the serum and ileum between the positive control group and high- dose treatment group ( P 〈 0.05, P 〈 0.01 ). Conclusion "Tongjie Oral Solution" caneffectively reduce the levels of IL-6 and TNF-c~ in both serum and ileum of rats with MODS reduced by lipopolysaccharide. The curative effect was markedly coincident with the dosage of the solution.
