黄芪多糖对粒细胞减少期侵袭性肺曲霉病SD大鼠血清半乳甘露聚糖的影响 被引量:2

Effect of astragalus polysacharin and amphotericin B treatment on serum galactaomannan levels in invasive pulmonary aspergillosis rats with granulocytopenia

摘要 目的 观察黄芪多糖(APS)与两性霉素B(AmB)联合应用对粒细胞减少期侵袭性肺曲霉病(IPA)大鼠血清半乳甘露聚糖(GM)值的影响,并探讨其作用机制。方法 40只清洁级雄性SD大鼠,随机分为5组,每组8只:(1)对照组:正常饲养;(2)模型组:建立IPA模型后正常饲养;(3)APS组:建模后腹腔注射APS 26.27mg(kg·d)×5d;(4)AmB组:建模后腹腔注射AmB 1.575mg(kg·d)×5d;(5)APS+AmB组:建模后分别予腹腔注射APS 26.27mg(kg·d)×5d+AmB 1.575mg(kg·d)×5d。(2)^(5)组大鼠经环磷酰胺(CT×)诱导、烟曲霉菌气管内接种建立粒细胞减少期大鼠IPA模型,并于接种后连续腹腔注射给药5d,第5天取大鼠动脉血以ELISA法检测血清中GM值,并取胸腺称重计算胸腺指数,根据统计分析结果评价APS对IPA模型大鼠胸腺指数和血清GM值的影响。结果 对照组胸腺指数高于其余4组,均有统计学差异(均P<0.05);APS组、APS+AmB组与模型组、AmB组比较胸腺指数升高,均有统计学差异(均P<0.05);模型组、APS组和APS+AmB组血清GM检测敏感性均为87.5%,AmB组为75.1%,对照组为0.1%;模型组、APS组、AmB组和APS+AmB组血清GM值升高,与对照组比较均有统计学差异(均P<0.05);APS+AmB组与APS组、模型组比较,血清GM值减低,均有统计学差异(均P<0.05)。结论 APS联合AmB治疗粒细胞减少期IPA大鼠能够减轻病情,减低血清GM值,其机制可能与APS提高胸腺免疫功能,减少烟曲霉菌菌丝数量有关。 Objective To investigate the effect of astragalus polysacharin (APS) and amphotericin B (AraB) treatment on serum galactaomannan (GM) levels in invasive pulmonary aspergillosis (IPA) rats with granulocytopenia. Methods Forty male Sprague-Dawley(SD) rats were randomly divided into five groups: control group, model group, APS group (intraperitoneal injec- tion of APS 26.27mg/kg·d), AraB group (intraperinoneal injection of AmB 1.575mg/kg·d) and APS+AmB group (intraperitoneal injection of APS 26.27mg/kg·d and AmB 1.575mg/kg·d). The IPA model was established by intratracheal inoculation of as- pergillus fumigatus after cyclophosphamide (CTX) induction in rats of groups 2, 3, 4 and 5. After 5 d of drug administration the arterial blood and thymus of rats were collected. The body and thymus were weighted and the thymus index was calculated; serum GM levels were measured by ELISA method. Results The thymus index in APS group and APS+AmB group was higher than that of AmB group and model group (P〈0.05). Serum GM levels in model group, APS group, Arab group and APS+AmB group were higher than that in control group (P〈0.05); and the serum GM level in APS+AmB group was lower than that in model group and APS group (P〈0.05). Conclusion The combination of APS and AmB can alleviate the disease extent and decrease serum GM level in rats with invasive pulmonary aspergillosis after granulocytopenia; which may be associated with the improve- ment of immune function of thymus.
