摘要 目的建立同时测定豨莶草中绿原酸和黄芩苷含量的方法。方法采用高效液相色谱-紫外检测法。色谱柱为Elite C18柱(200 mm×4.6 mm,5μm),流动相为体积分数0.1%磷酸水溶液-乙腈,梯度洗脱,流速为1.0 m L·min^-1,柱温为30℃,检测波长为280 nm。结果绿原酸和黄芩苷质量浓度分别在3.840~23.04 mg·L^-1(r=0.999 9)和0.480~2.88 mg·L^-1(r=0.999 7)内与峰面积呈良好的线性关系,平均回收率分别为99.9%和101.1%,RSD均为1.7%。结论该法专属、简便、准确,可用于同时测定豨莶草中绿原酸和黄芩苷的含量,为全面评价豨莶草药材质量提供了依据。 Objective To develop a new method for simultaneous determination of chlorogenic acid and ba- icalin in Siegesbeckia pubescens Makino. Methods HPLC-UV method was used to determination chlorogenic acid and baicalin. The separation was performed on a Elite C18 column (200 mm × 4.6 mm, 5μm)with 0. 1% phosphoric acid aqueous (A)- acetonitrile (B)as the mobile phase at the flow rate of 1.0 mL·min^- 1 The column temperature was 30 ℃ and the detection wavelength was 280 nm. Results The linear ranges of chlorogenic acid and baicalin were 3. 840- 23.04 mg· L^-1 ( r = 0. 999 9 ) and 0. 480- 2.88 mg·L^-1 ( r = 0. 999 7 ), respectively. The average recoveries were 99.9% and 101.1%, and their RSDs were both 1.7%. Conclusions This method is selective, simple and accurate, which can be applied for simultaneous determina- tion of chlorogenic acid and baicalin in Siegesbeckia pubescens Makino.
