大鼠牙髓干细胞的分离培养与鉴定 被引量:1

摘要 目的:分离培养大鼠牙髓干细胞并对其进行生物学鉴定。方法:采用酶消化法分离获得大鼠牙髓干细胞,有限稀释法纯化细胞,测定细胞克隆形成率,细胞计数法测生长曲线,抗波形丝蛋白及角蛋白、抗STRO-1免疫化学染色。体外分化诱导实验检测细胞的多向分化能力。结果:分离纯化获得的大鼠牙髓干细胞在体外具有一定的克隆形成能力,波形丝蛋白、STRO-1均有表达。诱导条件下部分牙髓干细胞具有多向分化的潜力,符合干细胞特征。结论:成功从大鼠牙髓中获取牙髓干细胞。 Objective:To isolate,culture and identify a rat dental pulp stem cell line.Methods:Rat dental pulp tissue was digested wit h mix ture of collagenase and the stem cells were isolated by limited dilut ion of culture cell for single cell clone.Drawing the growth curve,and observed the immunohistochemistry staining of vimentin,keratin and STRO-1.Differentiation potential of rat dental pulp was evaluated.Results:Isolated rat dental pulp stem cells have a certain ability to form clones in vitro,Vimentin,STRO-1 were all positive expression.Rat dental pulp stem cell have multilineage differentiation potential under inducing conditions,it accords with the characteristic of stem cells.Conclusion:Cells isolated from rat dental pulp are clonogenic,and have multipotent differentiation potential,satisfying the criteria of postnatal somatic stem cell.
