红霉素对烟草烟雾暴露致肺气肿大鼠肺血管超微结构的影响 被引量:1

摘要 目的:观察烟草烟雾暴露致肺气肿大鼠肺血管超微结构改变特点及使用红霉素(EM)干预的影响。方法:24只健康雄性Wistar大鼠随机分成对照组(A组)、烟草烟雾暴露组(B组)、EM预防组(C组),红霉素治疗组(D组),每组6只。A组予伪烟草烟雾暴露,B,C,D组于第1和第14天予气道内注入脂多糖(LPS),烟草烟雾暴露2h/d,持续8周制备模型;除第1天和第14天,C组每日EM灌胃,持续8周;D组烟草烟雾暴露4周后开始每日EM灌胃,持续4周;A、B组每日予等量的生理盐水灌胃,持续8周。对各组大鼠肺组织病理进行肺气肿和肺血管及其超微结构评估。结果:电镜下B组毛细血管内皮细胞线粒体减少、水肿,饮泡增多,与基底膜连接处增厚,肺小动脉内弹力膜菲薄,断裂,管壁大量胶原纤维增生。C组预防性干预超微结构上肺小动脉及毛细血管可见正常线粒体,内皮细胞无明显脱落,管壁平滑肌增殖但胶原纤维增生明显减少。D组毛细血管内皮细胞部分线粒体肿胀,基底膜连接处增厚,管壁可见增生的胶原纤维。结论:烟草烟雾暴露及LPS可引起大鼠肺血管内皮细胞损伤、血管壁平滑肌及胶原纤维增殖而致血管重构,早期使用EM干预可改善肺血管重构。
