摘要
目的探讨针灸预处理联合骨髓间充质干细胞(BMSCs)移植对心肌梗死大鼠心肌重构的影响。方法建立心肌梗死模型,将60只大鼠分成建立心肌梗死组,BMSCs组及干预组,每组各20只。BMSCs组及干预组在建模后24 h通过尾静脉注射1 ml BMSCs悬浮液。此外,干预组在建模前连续电针大鼠"内关"穴7 d,每次持续20 min,1次/d。建模4周后检测所有大鼠的前壁厚度、后壁厚度、左心室收缩末期内径(LVDs)、左心室舒张末期内径(LVDd)、左室射血分数(LVEF)及短轴缩短率(FS)指标。计算大鼠心肌梗死面积及梗死心肌区新生毛细血管密度。结果与心肌梗死组比较,BMSCs组以及干预组大鼠前壁厚度、FS、LVEF、毛细血管密度均明显升高,而LVDs、LVDd、心肌梗死面积明显下降,且干预组较BMSCs组更为明显(P均<0.05)。结论针灸预处理联合BMSCs移植可以改善心功能,减少心肌梗死面积,增加毛细血管数量,从而更有利于减缓大鼠心肌梗死区的心肌重构。
Objective To investigate the effects of acupuncture combined with hone marrow mesenchymal stem cells (BMSCs) on myocardial remodeling in rats after myocardial infarction (MI). Methods Totally 60 rats were established the myocardial infarction model and divided into the MI group, BMSCs group and experiment group, 20 rats in each group. The rats in the BMSCs group and experiment group were injected 1 ml BMSCs by caudal vein after 24 h of modeling, and the rats in the experiment group were given acupuncture for 7 d before the modeling additionally (each time for 20 min, once a day). The anterior and back wall thickness, end diastolic diameter (LVDs), left ventricular end diastolic diameter (LVDd), left ventricular ejection fraction (LVEF) and fractional shortening (FS) were detected by ultrasonic. The myocardial infarct area and infarcted myocardial capillaries density were detected and compared. Results Compared with the MI group, the anterior wall was thicker, FS, LVEF and capillary density were higher, and the levels of LVDs, LVDd and myocardial infarction area were obviously lower in the BMSCs and experiment groups (all P 〈 0.05). Meanwhile, the above indicators in the experiment group all showed significant differences as compared with the BMSCs group (all P 〈 0.05). Conclusion Acupuncture combined with bone marrow mesenehymal stem cells can improve cardiac function, reduce the myocardial infarction area, increase the number of capillaries,and thereby slow down the myocardial remodeling after infarction in rats.
