摘要
目的:探讨小檗碱对耐甲氧西林金黄色葡萄球菌(Methicillin-Resistance Staphylococcus aureus,MRSA)脂磷壁酸(Lipoteichoic acid,LTA)的释放作用。方法:采用微量肉汤稀释法测定小檗碱对临床分离的MRSA的最低抑菌浓度(Minimum Inhibitory Concentration,MIC)。酶联免疫法测定MRSA经小檗碱处理后的LTA含量,设置TritonX-100为阳性对照组,阴性对照组不加药。结果:小檗碱对MRSA分离株的MIC值为32μg·ml-1。随着培养时间的增长,各组细菌释放的LTA含量均有上升。与0.02%的Triton X-100相比,8μg·ml-1的小檗碱在各时间点对金黄色葡萄球菌LTA的促释放作用更强。结论:小檗碱可明显促进MRSA的LTA释放,该作用可能与其改变MRSA对β-内酰胺类抗生素耐药性有关。
Objective.. To investigate the effect of of berberine on LTA release from methicillin-resistance Staphylococcus aureus. Methods: Micro-broth dilution method was used to determined the minimum inhibitory concentration(MIC) of berberine against MRSA. The enzyme-linked irnmunosorbent assay(ELISA) was performed to detect the content of LTA from MRSA affected by berberine. TritonX-100 was used in place of bererine in positive control group. No drug was used in negative control group. Results.. The MIC of berberine against MRSA is 32μg · ml^-1. The content of LTA released from MRSA increased over time in each group. In camparison with 0. 02% Triton X-100, MRSA released higher level of LTA in the presence of 8 μg · ml^-1 berberine. Conclusion. Berberine is able to stimulate the release of LTA from MRSA, which may be related with the berberine's modification on antibacterial susceptibilty of β-lactams agaisnt MRSA.
