人工饲养大鲵皮胶原蛋白肽提取工艺研究 被引量:8

摘要 对6种蛋白酶进行筛选,优选出大鲵皮明胶酶解制备小分子胶原蛋白肽的三种最佳用酶为中性蛋白酶、木瓜蛋白酶及风味蛋白酶,添加量分别为木瓜蛋白酶8 000U/g,中性蛋白酶3 000U/g、风味蛋白酶600U/g。实验采用复合酶解工艺方法,以蛋白质回收率为参考指标,确定了最佳工艺参数:酶解时间为7h,酶解温度为55℃,料液比为1∶4。此时蛋白质的回收率为78.9%,其中相对分子量≤5 000Da占46.86%。 Through selecting from six enzymes,neutral protease,papain and flavourzyme enzymes were prepared small collagen peptide in Andrias davidianus skin. The addition of papain,neutral protease and flavourzyme was 8000U / g,3000U / g and 600U / g,respectively. Composite enzyme hydrolysis method was taken in this paper. The protein recovery was taken as reference index. The optimum technology parameter was defined. The result showed that enzymolysis time 7h,enzymolysis temperature 55℃,solid- liquid ratio 4: 1,the recovery rate of protein was 78. 9% and the relative molecular weight below 5000Da was 46. 86%.
