林蛙皮中胶原蛋白的提取工艺研究

摘要 目的采用酸酶法从林蛙皮中提取胶原蛋白。方法考察了药材粉碎度、加水量、酶的种类对林蛙皮中胶原蛋白提取率的影响。单因素考察结合正交实验得到胶原蛋白的最优提取条件。结果胶原蛋白的最优提取条件为:加水量为60 m L,提取时间为72 h,提取温度为37℃,加酶量为1%(0.05 g),加酸量为0.8 mol/L。结论此条件下林蛙皮中胶原蛋白的产率可达67.09%,并且此种提取胶原蛋白的方法操作简单、提取率高。 Objective To extract collagen from the skin of Rana chensinensis by acid enzyme method. Methods The effect of grinding degree of material,quantity of water and enzyme types on extraction rate of collagen in the skin of Rana chensinensis was investigated. The optimal conditions for the extraction of collagen were achieved by the combination of single factor investigation and orthogonal experiment. Results The optimal extraction conditions of collagen are the amount of water: 60 m L,extraction time: 72 h,extraction temperature: 37 ℃,the amount of enzyme: 1%( 0. 05g),acid: 0. 8 mol / L. Conclusion The yield of collagen in the skin of forest frog is 67. 09% under this condition.What's more,the extraction technology of collagen is simple with higher extraction rate.
