摘要
目的研究羊膜载体复合膜负载骨髓间充质干细胞在促进创面愈合中的作用。方法取健康纯种成年新西兰家兔30只,以羊膜载体复合膜为载体,将自体来源骨髓间充质干细胞植入家兔背部左侧的全层皮肤缺损创面为观察组,另仅以羊膜载体复合膜植入右侧的皮肤缺损创面为对照组,对比观察两组的术后创面愈合时间及病理和Brd U免疫组织化学染色检测等。结果观察组创面收缩率显著优于对照组,P<0.05,差异具有统计学意义。观察组术后4周的病理切片显示表皮细胞数目显著增多,表皮层显著增厚;免疫组织化学染色检测,可见染色阳性细胞,与周围表皮细胞无明显差异。结论羊膜载体复合膜负载骨髓间充质干细胞可显著提高创面的愈合质量,为皮肤损伤的完美愈合提供了治疗的新思路。
Objective To study the effect of amniotic membrane carrier composite membrane for loading bone marrow mesenchymal stem cells in promoting wound healing. Methods 30 healthy purebred adult New Zealand rabbits were selected as the objects. We applied amniotic membrane composite membrane as the carrier, and implanted the autologous bone marrow mesenchymal stem cells into full-thickness skin defect wound of rabbits’ left back. It was regarded as observation group. We only applied amniotic membrane carrier composite membrane to implant the skin defect wound of rabbits’ right back. It was regarded control group. The postoperative wound healing time and pathology and BrdU immunohistochemical staining detection were compared in two groups. Results In observation group, the wound shrinkage rate was signiifcantly better than control group, P〈0.05, there was statistical significance. In observation group after 4 weeks, the pathological biopsy showed that the number of epidermal cells significant increased, the epidermal layer was markedly thickened. After immunohistochemical staining detection, there were visible staining positive cells, there was no obvious difference with the surrounding epithelial cells. Conclusions The amniotic membrane carrier composite membrane for loading bone marrow mesenchymal stem cells can promote the quality of wound healing. It provided a new idea for perfect healing of skin damage.
