摘要
对鼠李上的白粉病进行显微观察,采用试剂盒法提取病原菌总基因组DNA,用PCR扩增其r DNA内转录间隔区(ITS)的保守序列,并通过Clustal等相关的生物信息学软件对其进行序列的比对和亲缘关系分析,在分子水平鉴定该菌种并比较与其它白粉菌菌种的亲缘关系。结果表明,本试验地发生的鼠李白粉病病原菌为费氏白粉菌,其与Gene Bank中Erysiphe friesii var.dahurica(登录号为AB000939)的序列相似度达99%。
Erysiphe sp. on Rhamnus davurica Pall was observed by microscope and its genome DNA was extracted for molecular identification and phylogenetic relationship analysis. The conserved se- quence of ITS (GenBank accession number is Banklt1743270) was amplified by PCR and aligned with other fifteen kinds of conserved sequence of powdery mildew, and the evolutionary tree was described and analysed. Morphological and molecular classification identification showed that the local powdery mildew pathogen was Erysiphe friesii which had the closest genetic relationship with E. friesii var. dahurica(ABO00939) from GeneBank,the sequence similarity was up to 99%.
