等离子体诱变选育解淀粉芽胞杆菌BI2高产抑菌物质

ARTP mutagenesis and screening of Bacillus amyloliquefaciens BI_2 producing high-yielding antimicrobial substances

摘要 解淀粉芽胞杆菌BI2是从青贮秸秆饲料中分离出的,对多种植物病原菌有较好抑制作用的菌株。文章旨在通过诱变筛选获得高产抑菌物质的菌株。利用常压室温等离子体(ARTP)对BI2菌株诱变处理,最终确定诱变处理的最佳时长为30 s,并经研究确定以黑曲霉作为指示菌,通过比较抑菌圈直径方法进行初筛,获得16株菌,然后经过发酵液抑制黄曲霉孢子萌发率,复筛出5株高产菌株,经10代连续培养,最终获得1株稳定高产菌株BI2-12,平均抑菌率比出发菌株高22.7%。 Bacillus amyloliquefaciens BL isolated from silage feed is an antagonistic strain, its secondary metabolites show a very good inhibitory effect on plant pathogens. This study aims to obtain a mutant strain producing more antimicrobial substances through ARTP mutation. The strain BL was used as original strain for Atmospheric and Room Temperature Plasma (ARTP) mutagenic treatment. The best treatment time for mutagenesis was finally determined as 30 s. Aspergillus niger as an indicator performance inhibition zone was used for primary screening, and 16 high-yield mutant strains were screened out according to the size of inhibition zones. Based on the inhibition rate level of spore germination of Aspergillus flavor, 5 high-yield mutant strains were rescreened out, and after 10 generations of continuous subculturing, a genetic stable and higb-yield mutant strain BI2-12 was obtained with 22.7% higher inhibition rate than the original strain. In conclusion, mutation of B. amyloliquefaciens using ARTP provides a new channel to develop the value of strains, a novel idea using inhibition zone to screen similar strains, as well as made good prospects for strains' development.
