肥胖小鼠血清铁调素水平变化及其与瘦素的关系 被引量:2

The level of serum hepcidin and its relationship with leptin in obese mice

摘要 目的:研究饮食诱导肥胖小鼠血清铁调素水平的变化及其与瘦素的相关性。方法:C57BL/6J小鼠随机分为正常对照组和肥胖组,每组6只。对照组饲以普通饲料,肥胖组以高脂饲料喂养12周。建模成功后小鼠心脏取血分离血清,用ELISA试剂盒测定血清中瘦素、铁调素的含量。结果:肥胖小鼠血清瘦素、铁调素含量显著高于正常对照小鼠[(33.92±9.39)μg·L-1vs(15.32±5.48)μg·L-1,P<0.01;(22.51±7.53)μg·L-1vs(14.03±1.08)μg·L-1,P<0.05];相关分析表明,血清铁调素与瘦素水平间呈显著正相关关系(r=0.917,P<0.05)。结论:肥胖小鼠血清铁调素水平升高,并且铁调素水平与瘦素浓度正相关。 Objective:To study the change in the concentration of hepcidin and its correlation with leptin in obese mice.Methods:C57BL/6J mice were randomly divided into two groups with 6 in each.Mice in the control group were put on a normal diet,and the obese group were fed with a high-fat diet to establish obese mice model.After 12 weeks of interventions,mice were sacrificed and blood samples were collected.Serum leptin and hepcidin contents were detected using enzyme-linked immunosorbent assay.Results:Compared with the control group,serum leptin level in the obese model group was significantly higher[(33.92 ± 9.39)μg · L^-1 vs (15.32 ±5.48)μg · L^-1,P < 0.01],serum hepcidin level in the model group increased significantly[(22.51 ± 7.53)μg ·L^-1 vs (14.03 ± 1.08)μg · L^-1,P <0.05] as well.Analysis suggested positive correlation between hepcidin and leptin levels(r =0.917,P <0.05).Conclusion:The concentrations of leptin and hepcidin in the serum of obese mice are significantly higher than in normal mice,and a positive correlation exists between leptin and hepcidin levels.
