隐丹参酮抑制A549细胞复发转移的机制 被引量:9

摘要 目的研究隐丹参酮对非小细胞肺癌A549细胞生长、迁移、侵袭、细胞周期分布的影响及机制。方法以不同浓度的隐丹参酮作用于体外培养的A549细胞,采用MTT法检测细胞增殖活性的改变;划痕实验和Transwell实验分析细胞迁移、侵袭能力的改变;流式细胞仪检测细胞周期及E-cadherin和β整合素的表达情况。结果 MTT检测结果显示,一定浓度的隐丹参酮可抑制A549细胞的增殖,且随着浓度的增加其抑制作用明显增强。划痕实验和Transwell实验显示,A549细胞经隐丹参酮作用后,其迁移和侵袭能力受到抑制。流式细胞术显示,A549细胞经隐丹参酮作用后,细胞周期阻滞于G1期,G1期细胞比例上升,S期细胞比例下降,且药物作用后细胞表面的E-cadherin表达上调、β整合素的表达下降。结论隐丹参酮可抑制非小细胞肺癌A549细胞的增殖、迁移、侵袭,其机制可能与E-cadherin的上调和β整合素下调及阻滞细胞周期有关。 OBJECTIVE To assess the effect of cryptotanshinone on cell growth , migration , invasion , and the cell cycle of non-small cell lung cancer cells.METHODS A549 cells were treated with different concentrations of cryptotanshinone in vitro .Cell proliferation was analyzed by MTT assay .Cell migration and invasion ability were eval-uated using wound scratch assay and transwell experiments .Flow cytometry was used to detect the change of cell cy-cle and the expression of E-cadherin and βintegrin.RESULTS The data from the MTT assay showed that crypto-tanshinone inhibited the proliferation of A 549 cells in an concentration dependent manner .Transwell experiments andwound scratch assay showed that cryptotanshinone could inhibit the migration and invasion ability of A 549 cells.Data from flow cytometry assay indicated that cryptotanshinone arrested cell cycle in G 1 phase ,rised proportion of cells in G1 phase and decreased proportion of cells in S phase .Furthermore,cryptotanshinone increased expression of E-cad-herin on the surface of cells ,while decreased expression of βintegrin.CONCLUSION Cryptotanshinone can inhib-it cell proliferation,migration and invasion ability of A549 cell.The mechanism may be related to cryptotanshinone effect on upregulation of E-cadherin ,downregulation of βintegrin and arrest of cell cycle .
