水稻草状矮化病毒侵染下水稻根发育相关基因的表达分析 被引量:1

Transcriptional Analysis of the Root Development Related Genes in Host Rice(Oryza Sativa)Infected with Rice Grassy Stunt Virus

摘要 水稻草状矮化病毒(Rice grassy stunt virus,RGSV)引起的水稻草矮病是热带和亚热带稻区的重要病害,RGSV侵染引起水稻植株根系严重发育不良。为解析RGSV侵染引致水稻根系发育不良的分子机制,以水稻品种‘汕优63’为材料,通过实时荧光定量RT-PCR技术分析水稻经RGSV侵染后根系发育相关基因Os PHR2、QHB、Os RR1、Os EXPB5、Crl-5和Os EXPA17等基因的m RNA表达量变化。结果表明,RGSV侵染引起Os PHR2和Crl-5基因表达下调,下调比率分别为2.18和4.86,而水稻病株QHB、Os RR1、Os EXPB5和Os EXPA17等基因的表达量与健株相比,各基因差异比率分别为1/1.49、1.01、1.39、1/1.37,表明RGSV侵染未引起这些基因的表达量发生变化。RGSV可能通过直接或间接调控根系发育相关基因表达水平而使根系发育异常。 Rice grassy stunt virus (RGSV) in tropical and subtropical areas can cause large losses in rice production and quality. RGSV-infected rice plants show root disorders and the molecular mechanism is still unclear. The transcript levels of root development related genes OsPHR2, QHB, OsRR1, OsEXPBS, Crl-5 and OsEXPA17 in rice plants infected with RGSV versus control rice plants were determined by using real-time quantitative reverse-transcription PCR. The results showed that RGSV infection led to reducing transcript levels of OsPHR2 and Crl-5 genes and the relative expressions reducing rate of OsPHR2 and Crl-5 genes were 2.18 and 4.86, respectively. RGSV infection did not alter the transcript levels of QHB, OsRR1, OsEXPB5 and OsEXPA17 genes whose relative expression levels were 1/1.49, 1.01, 1.39, 1/1.37, respectively. These data suggested that RGSV could cause root disorders by regulating transcription levels of root development related genes directly or indirectly.
