番茄红素对转基因果蝇模型神经保护作用的研究 被引量:3

摘要 目的研究番茄红素对β-淀粉样蛋白(Aβ)42转基因果蝇的神经保护作用。方法将实验果蝇随机分为不同浓度干预组、二甲基亚砜(DMSO)溶剂对照组、模型组和正常果蝇对照组,喂养至果蝇死亡。记录各组果蝇的寿命和运动爬行的结果以及测定果蝇体内超氧化物歧化酶(SOD)、胆碱酯酶活性及脑组织病理变化。结果番茄红素能延长Aβ42转基因果蝇的寿命,改善它的爬行能力,能增强其体内的SOD活性,胆碱酯酶活性受到抑制。同时番茄红素能减少模型果蝇脑组织病变空泡形成。结论番茄红素对Aβ42转基因果蝇具有神经保护作用,其机制可能是通过抗氧化作用来实现的。 [Objective] To study the neuroprotective effects of lycopene on the Aβ42 transgenic drosophila model of Alzheimer's disease. [Methods] Drosophila were randomly divided into four groups: normal control, normal drosophila exposed to DMSO, AI342 transgenie drosophila and Aβ42 transgenic drosophila exposed to lycopene of different concentrations (2.5, 7.5, 22.5 μg/g). The drosophila were fed until dead. The life span and the climbing ability of Drosophila were recorded and the superoxide dismutase (SOD) activity and the activity of AChE were determined. Made pathological sections to observe the pathological changes of brains in AD flies. [Results] Compared with Aβ42 transgenic drosophila group,the climbing ability and life span of Aβ42 transgenie drosophila applied with lycopene group were significantly improved. SOD activities of Aβ42 flies were increased and activity of AChE of Aβ42 flies were decreased significantly in LP groups than those in control group. Moreover, lyeopene can dramatically decreased brain vacuolization in Aβ42 flies. [Conclusion] Lycopene plays a significant role on the neuroprotective effects of Aβ42 transgenie drosophila. The mechanism by which lycopene rescue Aβ42 flies might be the antioxidant effect of lyeopene.
