摘要
目的探讨Bach2在胰腺癌中的表达及意义。方法采用逆转录-聚合酶链反应(RT-PCR)法检测胰腺癌(37例)、癌旁正常组织(29例)、人胰腺癌细胞株(3株)和正常导管上皮细胞株中Bach2的表达,采用蛋白质免疫印迹法和免疫组织化学(IHC)方法分别检测胰腺癌冰冻组织(18例)和石蜡包埋组织(15例)中Bach2的表达。结果 Bach2在胰腺癌表达明显下降(6.264±2.148)(P<0.05),在人胰腺癌细胞株BXPC-3、PANC-1和SWl990中表达明显下降(2.705±0.473)、(2.481±0.381)和(4.629±0.592)(P<0.01);胰腺癌组织和细胞株中Bach2蛋白质表达相对下降;胰腺癌神经侵犯者癌组织中Bach2表达更低(4.891±2.664)(P<0.05)。结论 Bach2在胰腺癌组织和细胞株中表达下降,可能与胰腺癌发生和神经侵犯相关。
[ Objective ] To investigate the expression of Bach2 in pancreatic cancer and its significance. [Meth- ods] The expression levels of Bach2 mRNA and in Pancreatic cancer(37cases), adiacent benign tissue(29 cases), 3 human pancreatic cancer cell lines and benignductal epithelial cell line were quantitated by using reverse transcrip- tase-polymerase chain reaction (RT-PCR). The protein expression of Bach2 was evaluated by Western blotting in 18 fresh frozen tissues and immunohistochemistry (IHC) in 15 paraffin-embedded specimens. [Results] Bach 2 was down-regulated significantly in pancreatic cancer (6.264+2.148), P 〈0.05) and cell lines BXPC-3, PANC-1 and SW1990(2.705+0.473), (2.481+0.381) and (4.629+0.592), P 〈0.01) respectively. The protein expression of Bach2 was obviously down-regulated in pancreatic cancer tissues and tumor cell lines. The group with perineural invasion showed lower expression of Bach2(4.891 +2.664), P 〈0.05). [ Conclusions ] The expression of Bach2 was down-regu- lated in pancreatic ductal adenocarcinoma and tumor cell lines, and this gene might be correlated to tumorigenesis and perineural invasion of pancreatic cancer.
