局部晚期鼻咽癌患者外周血VEGF检测及意义

Detection and Significance of VEGF in Peripheral Blood of Patients with Locally Advanced Nasopharyngeal Carcinoma

目的:研究血管内皮生长因子(vascular endothelium growth factor,VEGF)在局部晚期鼻咽癌患者外周血中的表达相,探讨VEGF与局部晚期鼻咽癌预后的关系,研究是否能预测局部晚期鼻咽癌患者的预后,或作为局部鼻咽癌治疗的靶分子,为寻找局部晚期鼻咽癌放化疗疗效的分子生物学评价指标提供依据。方法:选择57例局部晚期鼻咽癌患者,收集外周血标本,酶联免疫吸附实验(enzym-linked immunosorbent assay,ELISA)检测外周血中VEGF蛋白水平。结合临床病理特征和随访资料进行分析。结果:(1)局部晚期鼻咽癌患者外周血VEGF表达水平均与复发率、远处转移率有关(P<0.05)。(2)局部晚期鼻咽癌患者外周血VEGF的表达水平与性别和年龄无关(P>0.05)。(3)局部晚期鼻咽癌患者外周血VEGF的表达水平与生存率有关(P<0.05)。结论:局部晚期鼻咽癌患者外周血中VEGF的表达水平对复发率、远处转移率和生存率有影响,提示VEGF在局部晚期鼻咽癌的侵袭转移中可能起协同作用;VEGF检测对筛选较高鼻咽癌转移风险的患者可能有更大的临床价值。

Objective： To study the expression of vascular endothetium growth factor （VEGF） in patients with locally advanced nasopharyngeal carcinoma （NPC）, and to investigate the relationship between VEGF and prognosis in locally advanced nasopharyngeal carcinoma, in order to provide the basis for the prediction of prognosis and evaluation of biological index of locally advanced NPC. Methods： 57 patients with locally advanced nasopharyngeal carcinoma were selected, and their peripheral blood samples were collected. The expression levels of VEGF were detected by enzyme-linked immunosorbent assay, and the data was analyzed combined with clinical pathological characteristics and follow-up data. Results： （1）The expression level of VEGF in peripheral blood of patients with locally advanced nasopharyngeal carcinoma had a correlation with the recurrence rate, distant metastasis and survival rate. （2） The expression level of VEGF in peripheral blood of patients with locally advanced nasopharyngeal carcinoma had no correlation with sex and age. （3） The expression level of VEGF in peripheral blood of patients with locally advanced nasopharyngeal carcinoma correlated with 5-year survival rate.（4）The expression of VEGF in peripheral blood of patients with locally advanced nasopharyngeal carcinoma was positively correlated （c=0.54,P〈0.05）. Conclusions： The expression levels of VEGF in peripheral blood of patients with locally advanced nasopharyngeal carcinoma were related to recurrence rate, distant metastasis rate and survival rate, suggesting that VEGF could play a role in in the invasion and metastasis of locally advanced NPC, and VEGF detection in peripheral blood could be valuable to predict the prognosis of this disease.
