槲皮素对矽尘致肺纤维化的干预作用及对Prxs蛋白的表达影响研究 被引量:4

摘要 目的探讨槲皮素对矽尘致肺纤维化的干预作用以及对Prxs蛋白的表达影响,为矽肺的临床治疗提供新的思路。方法选取24只雄性健康成年SPF级SD大鼠,随机分为3组(对照组、染尘组和干预组)。对照组大鼠经气管注入无菌生理盐水1 ml,染尘组大鼠经气管注入50 mg/ml二氧化硅混悬液1 ml,干预组大鼠在气管灌注粉尘24 h后,以50 mg/kg的槲皮素每天灌胃进行干预,对照组和染尘组灌胃等量的无菌生理盐水。染毒28 d后处死大鼠。检测肺组织中H2O2和羟脯氨酸(HYP)的含量以及过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)的活性。采用Western blot方法检测抗氧化蛋白PrxⅠ和Ⅵ在肺组织中的表达水平,同时进行肺组织病理观察。结果矽尘染毒后肺组织中H2O2及HYP的含量升高,CAT及GSH-Px的活性降低;应用槲皮素干预后肺组织中H2O2及HYP的含量降低,CAT及GSH-Px的活性升高。染尘组与对照组相比,肺组织中PrxⅠ和Ⅵ蛋白表达含量分别降低了30.00%和18.00%,干预组与染尘组相比PrxⅠ和Ⅵ蛋白表达含量明显升高了34.29%、15.85%,差异均有统计学意义(P<0.05)。结论槲皮素对矽尘所致的肺纤维化有一定的保护作用。
