从细胞因子风暴探讨热毒宁注射液抗大鼠急性肺损伤作用机制 被引量:49

摘要 目的通过细胞因子风暴探讨热毒宁注射液对脂多糖(LPS)致大鼠急性肺损伤(ALI)的治疗作用及机制。方法采用LPS按6 mg/kg对SD大鼠气管内滴注制备ALI模型,观察热毒宁注射液3个剂量(10.16、5.08、2.54 g/kg)组和阳性对照药地塞米松注射液(0.5 mg/kg)iv给药后对大鼠肺组织匀浆白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)、IL-4、IL-6、IL-8、IL-10、细胞间黏附分子-1(ICAM-1)、核转录因子-κB(NF-κB)水平及肺组织形态学的影响。结果热毒宁注射液能够降低ALI模型大鼠肺脏IL-1β、TNF-α、IL-4、IL-6、IL-8、IL-10、ICAM-1、NF-κB水平,改善大鼠肺组织形态学变化。结论热毒宁注射液可改善肺组织病理变化,减轻LPS致ALI过程中炎性介质的生成和释放可能是治疗ALI的重要机制。 Objective:To investigate the effect and mechanism of Reduning Injection on the rats with acute lung injury(ALI) induced by lipopolysaccharide(LPS) from cytokine storm.Methods The model of ALI was established by instilling LPS(6 mg/kg) to SD rats.The effects of iv administration of Reduning Injection with three doses(10.16,5.08,and 2.54 g/kg) and control drug Dexamethasone(0.5 mg/kg) on IL-1β,TNF-α,IL-4,IL-6,IL-8,IL-10,ICAM-1,and NF-KB in lung homogenate and lung tissue pathology were observed.Results Reduning Injection could reduce the levels of IL-1β,TNF-α,IL-4,IL-6,IL-8,IL-10,ICAM-1,and NF-KB in lung tissue of rats,and improve the morphologic changes of lung tissue.Conclusion Reduning Injection could improve the pathological changes of lung tissue,and the important curative mechanism may be reducing the production and release of inflammatory mediators in ALI process induced by LPS.
