摘要
以拟南芥(Arabidopsis thaliana)Col生态型种子为材料,经EMS诱变获得一株后代表型分离比异常的突变体gpl1(group less 1,gpl1),进一步观察发现,其果荚短小萎缩,不能产生种子,花粉败育,成熟花粉粒无细胞核.另外,F1代的表型分析显示gpl1突变体的性状是受隐性基因控制,而F2代的遗传分离比分析显示gpl1突变体不遵循经典的遗传分离比定律.对gpl1基因进行图位克隆,并将其定位于2号染色体上T9J22与F12C20两个分子标记之间202kb的区段内.
Through EMS mutagenesis,the author obtained an Arabidopsis thaliana male sterile mutant gpl1 resulting in abnormal separation ratio,showing the reduced and unextended silique without seed and the sterile pollen without nuclear in microgainetophyte.In addition,phenotypes analysis of heterozygote F1 generation showed that the mutant was controlled by a recessive nuclear gene,however genetic separation ratio analysis of F2 generation showed that the mutant did not follow the classical genetic segregation ratio.The gpl1 gene was mapped to a region of 202 kb between the molecular makers T9J22 and F12C20 on chromosome 2 using map based cloning technique.
