摘要 旨在获得牦牛TLR10基因序列并进行生物信息学分析,同时分析其组织表达谱,为进一步研究牦牛TLR10基因的功能提供资料。以GenBank中牛TLR10基因序列设计引物,在牦牛脾组织中克隆出牦牛TLR10基因,应用半定量RT-PCR技术检测该基因组织表达情况。生物信息学分析表明,牦牛TLR10基因编码区全长2 328bp,编码氨基酸776个,分子质量196.3ku,理论等电点为4.94;TLR10编码的蛋白整体带负电荷,并表现为亲水性;进化树与同源性分析表明,牦牛与黄牛的同源性最高,达到99.4%,与水牛、绵羊、梅花鹿等哺乳动物遗传距离很近。组织表达谱显示牦牛TLR10基因在12个组织中均有表达。结果显示,牦牛TLR10基因高度保守,具有稳定的抗原特征。 The aim of the study was to clone TLR10 gene of yak, carry out bioinformaties analysis, assay gene expression profiling in different tissues and provide the basis for the further study in TLR10 gene of yak. This study designed primers and cloned TLR10 gene from yak spleen tissue as bovine TLR10 gene sequences in the GenBank, and the tissue specificity of TLR10 gene was analyzed by semi:quantitative RTPCR. The results of bioinformatics analysis showed that the length of the cloning TLR10 CDS is 2 328 bp. It encodes a peptide of 776 amino acids whose calculated molecular weight is 196.3 ku and PI is 4.94. The results of protein prediction showed that TLR10 protein is hydrophilic. Phylogenetic tree and sequence homology analysis showed that yak has higher homology with Bos taurus (99.4%) and also showed highly homology with cattle, sheep and other mammals. The tissue expression profile showed that the TLR10 gene was expressed in 12 tissues of yak. The results showed the TLR10 gene of yak is highly homological and characterized by stable antigen.
