摘要
[目的]优化黑木耳多糖酶法提取的工艺条件。[方法]采用复合酶法提取木耳多糖,以多糖的提取率为指标,考察了酶解温度、酶解溶液p H、浸提料液比以及酶解时间对黑木耳多糖提取效率的影响,并采用TLC薄层色谱层析法分析提取出的黑木耳多糖的成分。[结果]试验确定了复合酶酶解提取黑木耳多糖的最佳工艺条件:浸提料液比1∶40 g/ml,酶解溶液p H 7.0,酶解温度40℃,酶解时间3.0h。在此条件下,黑木耳多糖的提取率为4.353%,所含有的单糖为D-葡萄糖。[结论]研究可为黑木耳多糖的提取提供参考依据。
[Objective] To optimize compound enzymatic hydrolysis of Jew's-ear's polysaccharides. [Method]With polysaccharides yield as indicator,effects of enzymatic temperature,solution p H,solid-liquid ratio,enzymatic time on yield of Jew's-ear's polysaccharides were investigated. TLC method was used to analyzed the compounds of Jew's-ear's polysaccharides. [Result] The experiments determine the optimum conditions for enzymatic hydrolysis of Jew's-ear's polysaccharides: leaching agent multiples is 1 to 40,p H is 7. 0,temperature is 40 ℃,time is 3. 0 h. Under this condition,Jew's-ear's polysaccharides extraction rate is 4. 353%. The monosaccharide composition of polysaccharides is D-glucose. [Conclusion] The study can provide reference basis for extraction of Jew's-ear's polysaccharides.
