摘要
目的:建立稳定的成年SD大鼠心肌细胞缺氧模型,并对心肌细胞内Ca2+动态变化进行测定。方法:用缺氧液灌流30 min模拟心肌缺氧环境,共聚焦显微镜下测定Ca2+动态变化。结果:缺氧导致心肌细胞静息钙水平升高,钙瞬变峰值降低,钙瞬变上升和下降时程延长,肌质网钙储量下降,钙泵功能降低。结论:缺氧导致心肌细胞钙超载,对心肌细胞钙瞬变有抑制作用。
Objective:To establish a hypoxia model using SD rat cardiomyocytes,and to investigate their calcium dynamics. Methods:Calcium transients were recorded by a confocal calcium imaging system from rat cardiac myocytes which were perfused with hypoxia liquid for 30 minutes to simulate the hypoxia conditions. Results:The hypoxia produced a substantial increase in diastolic[Ca^2+]iand reduced the amplitude of calcium transients. Both rising time and decay time of Ca^2+ transients were significantly prolonged. The calcium content of sarcoplasmic reticulum(SR)decrease and function of calcium pump of SR reduce. Conclusion:The hypoxia can lead to cardiac myocytes calcium overload and inhibit calcium transients.
