摘要
目的:研究姜黄素在抑制异位子宫内膜细胞增殖过程中的影响,并且探讨该效果是否通过降低雌二醇(estradiol,E2)的生成所介导。方法:对异位内膜间质细胞、正常子宫内膜间质细胞、异位内膜腺上皮细胞和正常子宫内膜腺上皮细胞进行分离与培养。测定各组E2分泌量和姜黄素干预对细胞增殖的影响。结果:电化学发光免疫法结果表明,异位内膜腺上皮细胞E2生成水平高于异位内膜间质细胞生成水平(P〈0.05),同时正常子宫内膜间质和腺上皮细胞E2生成水平都极低。WST-8结果显示,与正常子宫内膜间质细胞相比较,异位内膜间质细胞生长速率更高。分别用10、30、50μmol/L浓度的姜黄素干预0-96 h后,异位内膜间质细胞的数量和生长速率比正常子宫内膜细胞减少;E2的生成水平比正常子宫内膜细胞低,尤其是在姜黄素浓度为30μmol/L和50μmol/L组中。结论:异位内膜局部雌激素高水平生成是异位内膜发生发展的重要因素,异位内膜腺上皮细胞在E2分泌中占主导地位。姜黄素能通过减少E2分泌来抑制异位子宫内膜细胞的增殖。
Objective To investigate the effects of curcumin on the proliferation of endometrial cells and whether the effect was mediated by down-regulating the production of estradiol ( E2 ). Methods Ectopic and eutopic endometrial interstitial cells, and glandular epithelium were isolated and cultured, respectively. The secretory quantities of E2 and the effects of curcumin on the proliferation of cells were determined. Results Electrochemiluminescence immunoassay results shewed that E2 level of ectopic endometrial glandular epithelium was significantly higher than that of eutopic endometfial glandular epitheli- um( P 〈 0.05 ), meanwhile, the expression of E2 in normal endometrial stromal and epithelial cells were extremely low. WST-8 result showed that ectopic endometriotic stromal cells had a higher growth rate than that of endometrial stromal cells. After treating with curcumin with a dose of 10,30 and 50 μmol/L for 0 to 96 h,the number and growth rate of endometriotic stromal cells was reduced, but the secretory quantities of E2 was lower compared with normal endometrial cells, especially in 30 μmol/L and 50 μmol/L curcumin group. Conclusion Local high level of E2 production in ectopic endometfial cells may paly important roles in the development of endometriosis, E2 was mainly secreted with ectopic endometrial glandular epithelium. Curcumin could inhibit the proliferation of ectopic endometrial cells through down-regulating the production of E2.
