摘要
目的:探讨安体舒通(Antisterone)对实验性糖尿病肾病(Diabetic nephropathy,DN)大鼠肾脏肝细胞生长因子(hepatocyte growth factor,HGF)的影响及其可能机制。方法:建立糖尿病肾病大鼠模型,随机分为糖尿病肾病组(DN组)和安体舒通治疗组(AT组),同时设正常对照组(NC组)。记录FBG、Scr及24 h尿微量白蛋白(24 h Upro)等指标。ELISA和Western blot法分别检测转化生长因子-β1(TGF-β1)、HGF的表达情况。结果:⑴DN组和AT组大鼠FBG、SCr、24 h Upro和RHI水平较NC组显著升高(P均<0.01),其中AT组Scr、24 h Upro和RHI均低于DN组(P<0.05,P<0.01);⑵DN组、AT组肾皮质TGF-β1蛋白表达较NC组显著升高(P<0.05),且AT组低于DN组(P<0.05);⑶DN组、AT组HGF表达量显著低于NC组(P<0.01),但AT组高于DN组(P<0.05)。结论:安体舒通可上调HGF的表达,减轻肾小球系膜区增生,延缓糖尿病大鼠肾脏病进展。
Objeotive To investigate the effect of antisterone on hepatocyte growth factor(HGF) in rats with diabetic nephropathy(DN) and its possible mechanism. Methods SD rats were randomly divided into 3 groups, the normal control (NC) group,diabetic nephropathy model(DN) group and diabetic nephropathy rats treated with antisterone(AT) group. Fasting blvod-glucose(FBG) ,24 h urinary protein (24 h Upro) and creatinine (Cr) were determined. The morphological changes of renal tissues were examined routinely by Masson staining. ELISA were used to determine the protein expression of TGF-β1 in renal tissue. HGF were measured by Western-Blot. Results Compared with group NC ,the levels of FBG, Scr, 24 h Upro and RHI were all obviously increased in both group DN and group AT( P 〈 0.01 ), while the levels of Scr,24 h Upro and RHI in group AT were significantly lower than those of group DN, ( P 〈 0.05 or P 〈 0.01 ). The level of TGF-β1 expression in group DN was significantly increased, and the effects could be abolished by antisterone ( P 〈 0.05 ). Furthermore, the level of HGF expression was significantly increased in group AT than that of group DN ( P 〈 0.05 ). Conclusion Antisterone could lighten the hyperplasia of mesangium and delay the progression of diabetic nephropathy in rats through upregulating HGF expression.
