摘要
目的 探讨顺铂 (DDP)对大鼠肾脏红细胞生成素 (Epo)产生、分泌的影响及与贫血的关系。 方法 经尾静脉注射DDP (8mg/kg)诱发大鼠贫血 ,采用胎肝细胞3 H TdR掺入法检测大鼠血清Epo的含量 ,并同时观察了大鼠肾组织在光镜及电镜下形态的改变。结果 在DDP诱发大鼠贫血过程中 ,血清Epo升高的幅度较小 ,血红蛋白 (Hb)降低持续的时间较长 ,血清尿素氮 (BUN)显著升高 ,病理切片证实DDP可引起大鼠明显的肾损伤。结论 DDP可抑制大鼠Epo的产生 ,Epo的缺乏是导致大鼠贫血的主要原因之一。
Objective To investigate effects of cis diamine dichloro platinum(DDP)on the production of serum erythropoietin(Epo) and secretion by the kidney and its association with anemia.Methods Anemia was induced in SD rats by DDP( 8 mg/kg ).Serum Epo levels were measured by 3 H TdR in fetal hepatocytes.Changes of renal tissue were observed by light microscope and electron microscope.Results The elevation of serum Epo was rather low.Lowering of hemoglobin concentration sustained a longer time in DDP induced anemia.Serum urea nitrogen elevated significantly.Pathological findings revealed that DDP might significantly cause renal damage in rats. Conclusions DDP might inhibit Epo production,which might be the main cause of anemia.
