抗ADAM_2抗体免疫磁珠捕获精子的新方法研究

Magnetic Bead-based Directional Capture of Sperm Using a Polyclonal Antibody Against ADAM_2

实验选用一种人类精子膜抗原对应的抗体-抗ADAM2制备免疫磁珠,对混合样本进行分离。将不同比例的精子与女性口腔上皮细胞混合,模拟制备出混合细胞悬液,加入实验制备的抗ADAM2抗体免疫磁珠反应,将分离出的细胞裂解,提取DNA,经毛细管电泳及图谱分析得到STR分型,将STR分型结果进行统计。实验过程只需在单管中完成,操作简便,对设备要求低,节省人力物力,实验制备的免疫磁珠为法医学处理性侵案件提供了一种新的技术手段,具有一定的应用前景。

This experiment prepared immunomagnetic beads with anti-ADAM2 antibody, use the immunomagnetic beads to capture the sperm from mixed sample.First, to extract DNA from the sperms isolated.Secondly, get the DNA-STR types by capillary electrophoresis and the DNA spectrum analyzer .Finally, statistic dates of the DNA typing result.This experiment is accomplished only in single pipe, easy to operate,has less strict to equipment,and save manpower material resources. So the immunomagnetic beads provide a new technical for solve the sexual sssault cases.So this method have broad prospects in forensic medicine.
