血浆样品中叶黄素的快速测定 被引量:2

摘要 目的建立一种血浆样品一步处理的简易方法,以解决血浆中叶黄素的大批量快速测定问题。方法取人体新鲜静脉血为样品,采用甲醇去蛋白并提取叶黄素,液相色谱测定。色谱条件为:C18色谱柱(250mmX4.60mm×5μm),甲醇:乙腈(65:35)为流动相,流速0.8ml/min,检测波长446nm,进样量20μl,柱温25℃。同时测定叶黄素标准溶液及血浆样品在常温下放置10h及一20oI=条件下放置10d内叶黄素含量的变化情况。结果叶黄素在0.005~0.200μg/ml浓度范围内与峰面积线性关系良好(r=0.999);回收率为95.2%~96.0%;方法的检出限(LOD)为0.010μg/ml,定量限(LOQ)为0.05μg/ml;相对标准偏差为1.5%。同时血浆样品在-20℃冷冻状态下10d之内稳定。结论该方法操作简单,稳定性好,适用于大样本人群叶黄素的测定。
