维拉帕米对甲状腺未分化癌耐药株阿霉素敏感性作用的研究 被引量:2

摘要 目的研究维拉帕米对甲状腺未分化癌耐药细胞株阿霉素敏感性的影响及其作用机制。方法用阿霉素诱导甲状腺未分化癌细胞株HTh74建立耐药细胞株HTh74R,RT—PCR检测HTh74和HTh74R细胞中耐药相关基因ABCG2、MDR1及MRP1的表达情况,四甲基偶氮唑盐微量酶反应比色法(MTY)检测阿霉素对HTh74、HTh74R、HTh74R联合维拉帕米组3组细胞增殖的影响,通过测定Caspase-3活性,探究维拉帕米的作用机制。结果HTh74R和HTh74细胞的形态及生长曲线无差异,但前者较后者具有显著增强的耐药性,且高表达ABCG2和MDR1基因,二者MRP1基因的表达无统计学差异;HTh74R细胞中,阿霉素联合维拉帕米组较阿霉素单药组半数抑制浓度(IC50)明显下降,且Caspase-3活性明显升高。结论ABC转运体阻滞剂维拉帕米通过提高Caspase-3的活性促进凋亡,增强HTh74R对阿霉素的敏感性,可能为解决甲状腺未分化癌临床药物治疗的耐药问题提供新的解决路径。
