目的了解前列腺素D2(prostaglandin D2,PGD2)-DPI受体途径相关的酶前列腺素G/H合成酶(cyclooxygenase,COX)-1、COX-2和脂质运载蛋白型前列腺素D合成酶(lipocalin—type prostagladin Dsynthase,L—PGDS)及DPI受体在人腹主动脉瘤(abdominal aortic aneurysm,AAA)组织中的表达定位,分析其与人AAA直径及破裂的相关性,探讨PGD2-DPI受体途径对人AAA的血管平滑肌细胞表型变异及炎性细胞趋化的影响。方法采用免疫组化及免疫荧光染色方法,检测已破裂、未破裂AAA组织和正常胸内动脉组织COX-1、COX-2、L—PGDS及DPI受体表达,利用RT—PCR方法检测人AAA组织中COX-1、COX-2、L—PGDS及DPI受体mRNA表达,使用Westernblot方法检测L—PGDS及DPI受体蛋白的表达,结合临床资料记录的AAA直径和是否破裂进行相关性分析。结果COX-1、COX-2、L—PGDS及DPI受体表达于人AAA组织的结构性细胞和炎性细胞,L—PGDS的表达量与AAA的直径和破裂呈负相关,COX-2的表达量与AAA的直径呈正相关。结论人AAA中存在PGD2-DPI受体途径相关的酶及受体表达,且COX-2的表达量与AAA直径相关,L—PGDS表达量与AAA直径和破裂相关。
Objective To map out the location of PGD2 - DP1 receptor pathways related enzymes, Cyclooxygenase(COX) - 1, COX - 2, L - PGDS and DP1 receptor expression in human abdominal aortic aneurysm (AAA) tissues, analyze its correlation with AAA diameter and ruptured. Methods The expression of COX - 1, COX - 2, L - PGDS and DP1 receptor in the ruptured and unruptured human AAA and mammary artery tissue were detected by immunohistochemistry in order to map out the expression location of these related enzymes and receptors in human AAA tissue. COX - 1, COX - 2,L - PGDS and DP1 receptor mRNA expression in human AAA were detected by RT - PCR, and L - PGDS and DP1 receptor protein expression were detected by Western blotting. We analyzed the correlation of AAA diameter and whether ruptured. Results COX - 1, COX - 2, L - PGDS and DP1 receptor express in AAA constitutive cells and inflammatory cells. The expression of L - PGDS was negatively correlated with the diameter and rupture of the human AAA, and the expression of COX - 2 was positively correlated with the diameter of the AAA. Conclusion There exist PGD2 - DP1 receptor pathways related enzymes expression in human AAA tissues. The expression quantity of COX - 2 correlates with AAA diameter, and L- PGDS expression correlates with AAA diameter and rupture.
